کنترل بیولوژیک بیماری بلاست برنج با استفاده از جدایه های بومی Trichoderma در استان مازندران
بیماری بلاست (Blast) با عامل Magnaporthe oryzae مهمترین بیماری برنج در جهان میباشد. استفاده از سموم شیمیایی اگر چه باعث کنترل بیماری میگردند، اما آسیبهای جدی متوجه انسان و محیط زیست میکنند. امروزه کنترل بیولوژیک بیماریهای گیاهی بهعنوان یک روش جایگزین و بیخطر، مورد توجه و تأکید ویژهای قرار گرفته است. در این تحقیق، نمونهبرداری از خاک شالیزار و اندامهای هوایی برنج در مناطق مختلف استان مازندران انجام شد و جدایههای Trichoderma با استفاده از محیط کشت انتخابی McFadden & Sutton’s RB-S-F جداسازی و خالص شدند. 89 جدایه تریکودرما با روشهای مختلف آزمایشگاهی علیه M. oryzae غربال شدند. در روش کشت متقابل، بیش از 90% جدایههای تریکودرما، موجب بازدارندگی رشد پرگنهی قارچ بیمارگر گردیدند. نتایج آزمایش هیپرپارازیتیسم نشان داد که هیفهای تریکودرما به دور هیفهای قارچ بیمارگر هیچ پیچشی نداشتند اما در بعضی موارد، جدا شدن هیفهای بیمارگر از محل دیوارهی عرضی مشاهده شد. در بررسی تأثیر متابولیتهای فرار، جدایههای تریکودرمای RP3-4 و RP3-2 بیشترین کاهش رشد بیمارگر را بهمیزان 6/66% سبب شدند. عصارهی کشت جدایهی RP7-1 در نسبت 20% تا 74/93% و در نسبت 5% تا 62/90% باعث کاهش رشد بیمارگر شد. براساس نتایج مطالعات آزمایشگاهی، تعداد 16 جدایه تریکودرما برای بررسی اثر آنها در کنترل بیماری بلاست در گلخانه انتخاب شدند. مطابق نتایج بهدست آمده از آزمونهای گلخانهای، جدایههای RP1-6 و RP4-2 بهترتیب با 100% و 99% کنترل بیماری، مؤثرترین جدایههای تریکودرما در کاهش شدت بیماری بلاست بودند. پس از این جدایهها،RP6-1 T. harzianum و RS7-6 T. virens بهترتیب با 98% و 97% درصد کاهش بیماری با قارچکش شیمیایی (کارپروپامید) در یک گروه آماری قرار گرفتند. براساس نتایج ارزیابی جمعیت جدایههای مختلف تریکودرما در پایان بررسی گلخانهای، میزان جمعیت جدایهها نسبتبه جمعیّت اولیه متغیر بوده است. جدایههایRP1-6 T. harzianum وT. atroviride RS4-1، بهترتیب بیشترین افزایش و کاهش جمعیّت را نسبت به جمعیّت اولیه بهکار رفته در گلخانه نشان دادند. ارتباطی بین دینامیک جمعیّت و کنترل بیماری مشاهده نگردید. همچنین، نتایج شناسایی 16 جدایهی تریکودرمای انتخابی با روشهای مورفولوژیکی و مولکولی نشان داد که 14، 1 و 1 جدایه بهترتیب متعلق به گونههای T. harzianum، T. virens و T. atroviride بودند.

بررسی نوسانات فصلی درصد پارازیتیسم پارازیتوئیدهای شبپرهی پشت الماسی، Plutella xylostella (Lep.: Plutellidae)در مزارع کلم گل جنوب تهران
شبپرهیپشتالماسی، Plutella xylostella مخربترین آفت گیاهان خانوادهی کروسیفر یا چلیپائیان (Brassicaceae) در سرتاسر دنیا میباشد. بهمنظور شناسایی و بررسی نوسانات جمعیّت پارازیتوئیدهای این آفت از خرداد ماه تا اوایل آبان ماه سال 1390 از مزارع کلم گل منطقهی جنوب تهران نمونهبرداری صورت گرفت. از هر مزرعه در مناطق جهان آباد، کهریزک، شکر آباد و پلائین قطعهای بهمساحت یک هکتار برای نمونه برداری انتخاب شد. در مزرعه تحقیقاتی دانشگاه شاهد نیز مزرعهای بهمساحت 600 متر مربع بهعنوان مزرعه بدون سمپاشی انتخاب شد. بوته بهعنوان واحد نمونه برداری در نظرگرفته شد و از هر مزرعه 20 بوته انتخاب شدند. نمونه برداری بهفواصل هر دو هفته یکبار انجام شده و تمام لارو و شفیرههای روی هر بوته جمعآوری گردید و در شرایط آزمایشگاهی و روی برگهای گیاه میزبان تا مرحلهی خروج پارازیتوئیدها پرورش داده شد. همچنین بهطریق فراخوانی پارازیتوئیدها جمعآوری شدند. در این بررسی سه گونه پارازیتوئید شناسایی شد که این گونهها عبارتاند ازDiadegma anurum (Thomson, 877)،Cotesia plutellae (Kurdjumov, 1912)، Oomyzus sokolowskii (Kurdjumov, 1912). در مجموع مناطق، بیشترین درصد پارازیتیسم توسط مجموع ارازیتوئیدها (75/47) در تاریخ 23 تیر ماه و کمترین آن (33/22) در تاریخ 29 خرداد ماه اتفاق افتاد. در مزرعه سمپاشی نشده بیشترین میانگین درصد پارازیتیسم در طول فصل (14/12) متعلق بهگونهی D. anurumبود. بیشترین درصد پارازیتیسم در مجموع (49/57)، در مزرعهی جهان آباد و کمترین درصد پارازیتیسم (22/8) در مزرعهی پلائین ثبت شد. نتایج این تحقیق نشان داد که درصد پارازیتیسم بین مزرعه سمپاشی نشده با سایر مزارع دارای اختلاف معنیداری است.

زهراگینی چهار جدایه از قارچ Beauveria bassiana روی جمعیّتهای تابستانگذران و زمستانگذران سن گندم، Eurygaster integriceps
در این تحقیق زهراگینی چهار جدایهی ایرانی Beauveria bassiana روی نمونههای تابستانگذران و زمستانگذران حشرات کامل سن گندم بهروش زیست سنجی از طریق اسپری کردن غلظتهای مختلف و نمونهبرداری در طول زمان، مورد بررسی قرار گرفت. سنهای کامل زمستانگذران حساسیّت بیشتری را نسبت بهسنهای کامل تابستانگذران نشان دادند (F= 69.99; df= 31, 1; P-value= 0.0001). مقادیر LC50 برای جدایهی فشند در مرحلهی تابستانگذران و مرحلهی زمستانگذران بهترتیب 108×1 و 105×2 اسپور بر میلیلیتر محاسبه شد. مقدار LC50 برای جدایهی Spt-566 روی سنهای زمستانگذران، 107×3 اسپور در میلیلیتر محاسبه شد. زمان لازم برای ایجاد مرگ و میر 50 درصد سنها (LT50) برای جدایهی فشند با 109 اسپور در میلیلیتر در دو مرحلهی رشدی سن تابستانگذران و زمستانگذران بهترتیب 13 و 7 روز و برای جدایهی Spt-566 در مرحلهی زمستانگذران 19 روز محاسبه گردید. جدایهی فشند با کمترین زمان لازم جهت مرگ و میر سنها و پایینترین LC50 بهعنوان جدایهی زهراگین مؤثر روی سن گندم تعیین شد. همچنین افزودن روغن به سوسپانسیون قارچ باعث افزایش شدت بیمارگری قارچ روی دو جمعیت سن گندم در همهی جدایهها شد.

ارزیابی کارآیی جدایههای Trichoderma و Bacillus در کنترل بیولوژیک پژمردگی فوزاریومی خربزه
در این تحقیق، کارآیی سه گونهی spp. Trichoderma و دو استرین باکتری Bacillus spp. در کنترل بیولوژیک Fusarium oxysporum f. sp. melonis عامل پژمردگی فوزاریومی خربزه در شرایط آزمایشگاه و گلخانه مورد ارزیابی قرارگرفت. آزمونها در قالب طرح کاملاً تصادفی در 4 تکرار انجام شد و برای مقایسهی میانگینها از آزمون LSD در سطح احتمال یک درصد استفاده شد. نتایج بررسیهای آزمایشگاهی روی کارآیی عوامل قارچی کنترل بیولوژیک نشان داد که در آزمون کشت متقابل به دو صورت قرارگیری همزمان و غیر همزمان عوامل کنترل بیولوژیک و بیماریزا، بهترتیب Trichoderma harzianum 6 و T. viride 10 با 70 و 59/52 درصد بیشترین میزان بازدارندگی را روی رشد پرگنهی عامل بیماریزا داشتهاند. همچنین، آزمون ترکیبات فرار آنها مشخص نمود که بیشترین میزان بازدارندگی رشد پرگنهی عامل بیماریزا (5/31 %) به T. harzianum 6اختصاص داشته است. از طرف دیگر، نتایج بررسیهای آزمایشگاهی روی کارآیی عوامل باکتریایی کنترل بیولوژیک نشان داد که در آزمون تقابل و تولید ترکیبات فرار، بیشترین میزان درصد بازدارندگی رشد پرگنهی عامل بیماریزا بهترتیب 5/30 و 37/12 درصد به Bacillus spp. 62 وBacillus spp. 15اختصاص داشته است. در بررسی ترکیبات مایع خارج سلولی قابل نفوذ در آگار، کلیهی استرینهای باکتری مورد مطالعه، مانع رشد پرگنهی عامل بیماریزا گردیدند. در آزمایشات گلخانهای مایهزنی گونههای تریکودرما و استرینهای باکتری بهصورت بذرمال استفاده شد. درصد گیاهان زنده در گلدانهایی که از استرینهای .Bacillus spp و مخلوط بین گونههای .Trichoderma spp تیمار شده بودند و همچنین شاهد بدون عامل بیماری 100 درصد بود. اما در سایر تیمارهای مورد استفاده نسبت به شاهد سالم از 2/65 تا 38/79 درصد متغیر بود.

بررسی دفاع سلولی لاروهای (Utethesia pulchella (Lepidoptera: Arctiidae در مقابل قارچهای Beauveria bassiana و Isaria farinosae
دفاع سلولی حشرات یکی از راهکارهای مهم آنها در برابر عوامل بیگانه مانند اسپور قارچها، باکتریها، نماتدها و تخم پارازیتوئیدها میباشد. با شناخت سیستم ایمنی حشره بهتر میتوان از روشهای کنترل میکروبی علیه آنها استفاده کرد. در این تحقیق، واکنش ایمنی سلولی لاروهای سن چهارم پروانه Utethesia pulchella L. در برابر دو جدایه از قارچ Beauveria bassiana (Bals.-Criy) شامل Fashand و 566و یک جدایه قارچ(Holmsk) Fr. Isaria farinosae بهنام 1872c بررسی شد. غلظت 105 اسپور در میلیلیتر از جدایههای قارچی بهلاروها تزریق شدند و فعالیت سلولهای خونی، گره زایی و آنزیم فنل اکسیداز پس از 1، 3، 6 و 12 ساعت ارزیابی شدند. تیمار شاهد شامل لاروهایی بود که با آب مقطر تزریق شدند. تعداد کل سلولها، تعداد پلاسموتوسیتها و گرانولوسیتها در زمانهای 3 و 6 ساعت پس از تزریق حداکثر بود که تفاوت معنیداری را با شاهد نشان داد. اما بهتدریج پس از 6 ساعت تعداد کل سلولها و تعداد افتراقی سلولهای خونی کاسته شد. پلاسموتوسیتها و گرانولوسیتها در فرایند گرهزایی شرکت کرده و منجر بهنابودی عامل بیگانه شدند. فرایندگرهزایی در فواصل زمانی 3 و 6 ساعت پس از تزریق عوامل بیگانه در همولنف حشره حداکثر بود. فعالیت فنل اکسیداز در حضور سوبسترای L-DOPA اندازهگیری شد. این پارامتر با تعداد سلولهای خونی در زمانهای مختلف ارتباط مستقیم نشان داد. بالاترین فعالیت این آنزیم 3 و 6 ساعت پس از تزریق بود. نتایج این بررسی نشان داد که لاروهای U. pulchella فعالیت دفاعی خوبی در برابر اسپورهای این 3 جدایه نشان دادند و توانستند با ایجاد گرهها اطراف عامل بیگانه آنها را نابود کنند.

بررسی خاصیت ضدقارچی نانو امولسیون اسانس برخی گیاهان دارویی در کنترل پوسیدگی نرم توتفرنگی ناشی از Rhizopus stolonifer
هرچند قابلیت برخی اسانسهای گیاهان دارویی بهعنوان قارچکش شناخته شده است اما ضمن دشواریهایی درکاربرد، دارای اثرات ارگانولپتیک (organoleptic) نیز بوده و در محصولات ایجاد بو و مزه میکنند. یکی از روشهای بهحداقل رساندن این معایب اسانسها، استفاده از نانوامولسیون آنهاست که ضمن پایداری باعث افزایش خواص ضدمیکروبی از طریق افزایش جذب سلولی میشود. در تحقیق حاضر، نانوامولسیون اسانس سه گیاه دارویی نعنافلفلی (Mentha piperita)، آویشن دنایی (Thymus daenensis) و مرزهی خوزستانی (Satureja khuzistanica) تهیه و اندازهی ذرات آنها مشخص شد. ترکیب اصلـی در اسـانسهای آویشن دنـایی و مرزهی خوزستانی بهترتیب شامل تیمول (8/64 درصد) و کارواکرول (4/72 درصد) بود در حالیکه منتول (5/36 درصد) و منتون (8/33 درصد)، مهمترین ترکیبات موجود در اسانس نعنافلفلی بودند. اسانسها و نانوامولسیون آنها بههمراه قارچکش تیابندازول در غلظتهای مختلف (یک، دو و چهار در هزار) در کنترل قارچ Rhizopus stolonifer، عامل پوسیدگی میوهی توتفرنگی مورد مطالعه قرارگرفتند. بهطورکلی، نانوامولسیون اسانسها خاصیت ضدقارچی بیشتری نسبتبه اسانسها داشتند. در غلظت چهار در هزار، اسانس و نانوامولسیون مرزهی خوزستانی پوسیدگی میوه را بهطور کامل متوقف کرد و فعالیت ضدقارچی بیشتری نسبت به قارچکش تیابندازول (83/85 درصد کنترل) نشان داد. بین تیابندازول با نانوامولسیون اسانسهای آویشن دنایی و نعنافلفلی تفاوت معنیدار در کاهش بیماری مشاهده نشد. در غلظت دو در هزار نیز نانوامولسیون مرزه با 50 درصد کنترل، تأثیر معنیداری در کاهش پوسیدگی نشان داد. در پایان، نانوامولسیون اسانسها بهخصوص اسانس مرزه برای ساخت قارچکشهای طبیعی پیشنهاد میشود.

ساخت فرمولاسیون سوسپانسیون غلیظ میکروکپسول باکتریBacillus thuringiensis
در این پژوهش تولید میکروکپسول باکتری باسیلوس تورینجینسیس با پلیمر سدیم آلژینات به روش امولسیون دوفازی مورد بررسی قرار گرفت. پایداری فرمولاسیون میکروکپسول تولید شده در برابر اشعهی ماورای بنفش با طول موج 400 و 254 نانومتر بررسی شد. نتایج آزمایشات نشان داد درصد زندهمانی اسپورها در فرمولاسیون میکروکپسول سدیم آلژینات 5% در برابر اشعهی ماورای بنفش با طول موج 400 نانومتر و 254 نانومتر بهترتیب 10 و 14% کاهش و در حالت غیر میکروکپسول در برابر طول موجهای مذکور 60 و 40% کاهش یافت. در آزمون زیست سنجی، درصد مرگ و میر لارو سن دوم افستیا بعد از 10 روز، برای فرمولاسیون میکروکپسول اشعه دیده با طول موج 400 نانومتر، 10% کاهش و در حالت غیرمیکروکپسول در برابر طول موج مذکور 78% کاهش داشت. قطر ذرات میکروکپسول توسط میکروسکوپ نوری در حدود 7-20 میکرومتر و بازدهی میکروکپسول 86% بهدست آمد.

گزارش زنبور Anagyrus novickyi (Hym.: Encyrtidae)، پارازیتویید شپشک آردآلود توت، Pseudococcus comstocki از ایران
بهمنظور مطالعهی دشمنان طبیعی شپشک آردآلود توت Pseudococcus comstocki (Kuwana) (Hem.: Pseudococcidae)، نمونهبرداریهایی در شهرستان تبریز (استان آذربایجان شرقی) انجام گرفت. در این تحقیق یک گونه زنبور پارازیتویید از خانوادهی Encyrtidae جمعآوری و تحت عنوان Anagyrus novickyi (Hoffer) شناسایی گردید. این گونه برای نخستینبار از ایران گزارش میشود.

گزارش میزبانی زنبور Hyposoter clausus (Ichneumonidae: Campopleginae) پارازیتویید لارو سفیدهی کوچک کلم،Pieris rapae از مزارع کلم گل تهران
بهمنظور شناسایی پارازیتوییدهای سفیدهی کوچک کلم، Pieris rapae (Linnaeus, 1758) (Lepidoptera: Pieridae) از مزارع کلمگل در جنوب تهران، در سال 1390 نمونهبرداری صورت گرفت. در بین نمونهها گونهی Hyposoter clausus (Brischke, 1880)(Hymenoptera: Ichneumonidae) برای اولینبار از از روی سفیدهی کوچک کلم و منطقهی تهران گزارش میشود. گونهی دیگر بهنام Cotesia glomerata L. (Hymenoptera: Braconidae) نیز به عنوان پارازیتویید این آفت جمعآوری و شناسایی شد.
