In this study, 60 strains of pseudomonad fluorescent bacteria were obtained and isolated from wheat rhizosphere in Tehran, Ghazvin, Golestan, Ardebil, Zanjan, Markazi and Esfahan provinces and their efficacy in biological control of F. culmorum was investigated. According to biochemical, physiological and morphological characteristics of the bacteria, the strains were very similar to biovars І, ІІІ and V of Pseudomonas fluorescence and P. protegens. Antagonistic activities of pseudomonad fluorescent strains against F. culmorum were then evaluatedResults showed that 11 isolates along with CHA0 were the most effective strains. In order to determine competition and colonization abilities of the selected stains in the soil and wheat rhizosphere, the strains were tested on four wheat cultivars for three continuous growth cycles, each which was 42 days long. The results indicated that Niknejad cv. had the highest bacterial population density after three cycles, followed by Zagross cv. The over all results of this study showed that the bacterial strains 19 and 27 (both belonging to P. progegens) had a very good antagonistic ability and have the potential for biological control of F. culmorum.  }, keywords = {Wheat,crown rot root,antagonism,Pseudomonas fluorescens,2,4-diacetylphloglucinol antibiotic gene (phlD)}, title_fa = {بررسی توانایی تعدادی از جدایه‌هایPseudomonas fluorescens در کنترل بیولوژیکی بیماری پوسیدگی طوقه و ریشه‌ی گندم (Fusarium culmorum)}, abstract_fa = {بیماری پوسیدگی فوزاریومی طوقه و ریشه یکی از مهمترین بیماری‌های گندم در دنیا و ایران می‌باشد که توسط چندین گونه فوزاریوم از جمله Fusarium culmorum و F. pseudograminearumایجاد می‌شود. در این تحقیق 60 نمونه باکتری از سودومونادهای فلورسنت از محیط فراریشه (rhizosphere) از مناطق گندم کاری استان‌های تهران، قزوین، گلستان، اردبیل، زنجان، مرکزی و اصفهان جمع‌آوری، جداسازی و اثر آن‌ها در شرایط آزمایشگاه و گلخانه در کنترل بیولوژیکی قارچF. culmorum  (عامل بیماری پوسیدگی ریشه‌ی گندم) مورد بررسی قرار گرفت. با توجه به آزمون‌های بیوشیمیایی، فیزیولوژیکی و مورفولوژیکی و داشتن ژن تولید کننده‌ی آنتی‌بیوتیک 2 و 4- دی استیل فلوروگلوسینول PhlD، ویژگی‌های جدایه‌های انتخاب شده با بیووارهای I, III, V گونه‌ی Pseudomonas fluorescensو P. protegensبیشترین شباهت را داشتند. نتایج بررسی اثرات بازدارندگی جدایه‌های سودوموناس روی قارچF. culmorum  (عامل بیماری پوسیدگی ریشه‌ی گندم) نشان داد که یازده جدایه به‌همراه جدایه‌یCHA0  (جدایه بین‌المللیCHA0  که اولین‌بار از روی توتون در سال 1992 توسط کیل و همکاران از سوئیس گزارش شده بود) بیشترین تأثیر را در جلوگیری از رشد قارچ نشان دادند. قدرت رقابت و کلونیزاسیون استرین‌های مختلف روی چهار رقم گندم شامل ارقام نیک نژاد، مرودشت، زاگرس و سرداری در سه سیکل متوالی رشد گندم بررسی شد. طول هر دوره معادل 42 روز تعیین گردید. براساس نتایج، رقم نیک نژاد بیشترین میزان جمعیت باکتری را نسبت به سایر ارقام در ریزوسفر خود تکثیر نموده و رقم زاگرس کمترین جمعیت باکتری را در ریشه‌ی خود داشت. نتایج بررسی‌های گلخانه‌ای نشان داد که ارقام مرودشت و زاگرس بیشترین شدت آلودگی در طوقه و ریشه و ارقام سرداری و نیک نژاد کمترین میزان آلودگی را نشان دادند و نسبت به بیماری متحمل‌تر بودند. براساس نتایج کلی این بررسی جدایه‌های 19 و 27 باکتری که هر دو به گونه‌ی P. protegens شباهت زیاد داشتند از توان آنتاگونیستی بسیار خوبی برخوردار بودند.}, keywords_fa = {گندم,پوسیدگی ریشه و طوقه,باکتری آنتاگونیست,سودوموناس فلورسنت,ژن PhlD}, url = {https://jbiocontrol.areeo.ac.ir/article_100605.html}, eprint = {https://jbiocontrol.areeo.ac.ir/article_100605_8e6942d3def9b9e1b90e5d75d0edf5b2.pdf} }
