Meloni, the causal agent ofFusarium wilt disease of melon in the laboratory and greenhouse conditions. Dual culture, antimicrobial metabolites, and volatile metabolites were used in In vitro assay. Fungal pathogen colony area was recorded, compared with control and induced inhibition of growth was determined. A greenhouse experiments was also performed to test the efficacy of Trichoderma and Bacillus isolates in biological control of Fusarium oxysporum f. sp. melonis. This experiment displayed the antagonistic ability using dual culture simultaneous and dual culture non-simultaneous assays against fungal pathogen. Results showed that Trichoderma harzianum 6with 70% and T. viride10 with 52.9 reduction in the growth of fungal pathogen were the most efficient isolates respectively in comparison with the control. Results of dual culture assays also indicated that Bacillus 62 with 30.5%, in volatile metabolite test, Bacillus 15whit 12.37% and cell free culture test of Bacillus with 100% , were the most efficient in reducing mycelia growth of pathogenic fungus. Percent survival of the melon plants in pots treated with a mixture of both Bacillus, and Trichoderma isolates and healthy control was 100. However, the number of survived plants in other treatments varied from 65.2 to 79.38%.  The above results were obtained statistically by mean comparison at 1% probability level using LSD test.}, keywords = {biocontrol,Fusarium Wilt,melon,Trichoderma spp,Bacillus spp}, title_fa = {ارزیابی کارآیی جدایه‌های Trichoderma و Bacillus در کنترل بیولوژیک پژمردگی فوزاریومی خربزه}, abstract_fa = {در این تحقیق، کارآیی سه گونه‌ی spp. Trichoderma و دو استرین باکتری Bacillus spp.  در کنترل بیولوژیک Fusarium oxysporum f. sp. melonis عامل پژمردگی فوزاریومی خربزه در شرایط آزمایشگاه و گلخانه مورد ارزیابی قرارگرفت. آزمون‌ها در قالب طرح کاملاً تصادفی در 4 تکرار انجام شد و برای مقایسه‌ی میانگین‌ها از آزمون LSD در سطح احتمال یک درصد استفاده شد. نتایج بررسی‌های آزمایشگاهی روی کارآیی عوامل قارچی کنترل بیولوژیک نشان داد که در آزمون کشت متقابل به دو صورت قرارگیری هم‌زمان و غیر هم‌زمان عوامل کنترل بیولوژیک و بیماری‌زا، به‌ترتیب Trichoderma harzianum 6 و T. viride 10 با 70 و 59/52 درصد بیشترین میزان بازدارندگی را روی رشد پرگنه‌ی عامل بیماری‌زا داشته‌اند. همچنین، آزمون ترکیبات فرار آن‌ها مشخص نمود که بیش‌ترین میزان بازدارندگی رشد پرگنه‌ی عامل بیماری‌زا (5/31 %) به T. harzianum 6اختصاص داشته است. از طرف دیگر، نتایج بررسی‌های آزمایشگاهی روی کارآیی عوامل باکتریایی کنترل بیولوژیک نشان داد که در آزمون تقابل و تولید ترکیبات فرار، بیش‌ترین میزان درصد بازدارندگی رشد پرگنه‌ی عامل بیماری‌زا به‌ترتیب 5/30 و 37/12 درصد به Bacillus spp. 62 وBacillus spp. 15اختصاص داشته است. در بررسی ترکیبات مایع خارج سلولی قابل نفوذ در آگار، کلیه‌ی استرین‌های باکتری مورد مطالعه، مانع رشد پرگنه‌ی عامل بیماری‌زا گردیدند. در آزمایشات گلخانه‌ای مایه‌زنی گونه‌های تریکودرما و استرین‌های باکتری به‌صورت بذرمال استفاده شد. درصد گیاهان زنده در گلدان‌هایی که از استرین‌های .Bacillus spp و مخلوط بین گونه‌های .Trichoderma spp تیمار شده بودند و همچنین شاهد بدون عامل بیماری 100 درصد بود. اما در سایر تیمارهای مورد استفاده نسبت به شاهد سالم از 2/65 تا 38/79 درصد متغیر بود.}, keywords_fa = {پژمردگی فوزاریومی,کنترل بیولوژیک,خربزه,Trichoderma spp,Bacillus spp}, url = {https://jbiocontrol.areeo.ac.ir/article_100335.html}, eprint = {https://jbiocontrol.areeo.ac.ir/article_100335_c91908d0195f9e72d1fa8dd9fbaaba13.pdf} }
